Polyacrylamide microcapsules have been investigated as carriers for achieving site specific delivery of 5-Fluorouracil (5-FU). Acrylamide was cross-linked with N N' methylene-bisacrylamide using an emulsion polymerisation procedure. Spherical microcapsules with mean diameter 2.65 +/- 0.37 mu m and drug entrapment effciency of 60.33 +/- 3.68 percent were obtained. In vivo organ distribution studies in rats showed maximum drug accumulation in lungs, followed by liver and kindneys. Time course vs. drug tissue concentration profiles indicate that the deposition of the drug in the organs occured at a slow rate, showing peak drug levels at 24 hours post injection.
