Development and Validation of a Rapid and Simple High Performance Liquid Chromatography Method for Determining Hydrocortisone

A new, selective, and sensitive high-performance liquid chromatography method was developed for measuring head circumference in bulk and at medicinal concentrations. The high-performance liquid chromatography analysis was performed using a Waters system equipped with a C18 (2500×4.6 mm) 5 µm column. The mobile phase consisted of a mixture of acetonitrile, methanol, and water in a 40:40:20 (v/v/v) ratio, with a flow rate of 1.0 ml/min. Hydrocortisone was retained for approximately 2 min and detected at a wavelength (gamma) of 254. Specificity, linearity, limit of detection, limit of quantification, accuracy, precision, and robustness were all validated to meet the international conference on harmonization criteria. All the results were satisfactory, indicating that the method is appropriate for its intended use in routine drug assays and quality control.
